Step 1-2: Vial Thaw and Inoculum Expansion
A vial of the Working Cell Bank (WCB; Lot 05BT03) is thawed in a warm water bath and
the cells are diluted with an appropriate amount of animal protein free (APF) Inoculum
expansion medium containing 500 nM MTX (methotrexate). Following centrifugation, the
cells are inoculated into a 250 mL shake flask containing approximately 75 mL of APF
Inoculum expansion medium with 500 nM MTX. The culture is subsequently expanded to
approximately 350 mL in a single 1 L shake flask. The next two expansion steps are
performed in a 20 L ***rocker bag*** (CultiBag RM). A final volume of approximately 9 L is used
to inoculate the 100 L seed bioreactor. The flasks are placed on a shaking platform at 120
RPM in an incubator at a controlled temperature of 37°C, 6% CO2. The rocker bag
bioreactor is operated at 19 rocks per minute, 0.25 standard liters per minute (SLPM) air
flow, temperature of 37°C, and 6% CO2. The duration of each stage is approximately 2 to 3
days. The cells are seeded between 4.0 × 105 and 5.0 × 105 viable cells/mL and expanded to
the next stage when viable cell density (VCD) reaches at least 2.0 to 2.8 × 106 viable
cells/mL, depending on the stage of the culture.
